Slipping chain clonk in first gear

Hi all - got something with my 3TB that I'm still not able to puzzle out.

When you pull away with a bit of poke in first I get a mechanical clunking and momentary loss of power. Feels like the front sprocket is slipping but I don't think this is the case. In second you can completely floor it. Any ideas?

Bike is 3TB 1991, bottom end rebuilt 10,000 miles ago so 1st gear should be fine. Chain and sprockets are DID and JT and were fitted about 4000 ago and don't look bad at all. I do keep my chain on the looser end of the spectrum, could it be some kind of transient bunching? Maybe I'll tighten it and see.

Clutch?
About 4,000 miles ago I had clutch slip due to using fully synth and have since swapped (same plates) to semi which has fixed. Doesn't feel like clutch slip though.

When I get round to it i'll jog along side while a mate gives it some gas but until then im going gentle!

Cheers

Tom
